MicroScale Thermophoresis – Example measurement

ERK2 (MAPK1, mitogen-activated protein kinase 1) is an important player in multiple biochemical signaling pathways and it is involved in different cancer types. In this example study, we analyzed the binding affinity of ERK2 to four different binding partners: a specific ERK2 inhibitor, an ERK2 inhibitor peptide, an aptamer specific for ERK2 and an anti-ERK2 antibody. The Kd of the four different molecular interactions was determined. The specific anti-ERK2 antibody presents the strongest binding affinity to ERK2. These results allow to determine the best binding partner for ERK2 within a temperature range of 20–45°C.
